LIF detection often requires labeling of analytes with fluorophores; and fast fluorescent derivatization is valuable for high‐throughput analysis with flow‐gated CE. Here, we report a fast fluorescein‐labeling scheme for amino acid neurotransmitters, which were then rapidly separated and detected in flow‐gated CE. This scheme was based on the reaction between primary amines and o‐phthalaldehyde in the presence of a fluorescent thiol, 2‐((5‐fluoresceinyl)aminocarbonyl)ethyl mercaptan (FACE‐SH). The short reaction time (<30 s) was suited for on‐line mixing and derivatization that was directly coupled with flow‐gated CE for rapid electrophoretic separation and sensitive LIF detection. To maintain the effective concentration of reactive FACE‐SH, Tris(2‐carboxyethyl)phosphine was added to the derivatization reagents to prevent thiol loss due to oxidation. This labeling scheme was applied to the detection of neurotransmitters by coupling in vitro microdialysis with online derivatization and flow‐gated CE. It is also anticipated that this fluorophore tagging scheme would be valuable for on‐chip labeling of proteins retained on support in SPE.  相似文献

Accumulation of chlorpyrifos (CP), a pesticide, causes a significant environmental problem in food, surface/ground waters further to human health. The removal of the CP pollutant in surface/wastewater could be achieved by biochar due to the improved physical and chemical properties. In this work, the CP removal capacities of biochar samples derived from walnut shells at various temperatures from 450 to 900 °C were investigated. The experiments were performed as laboratory batch type study and the adsorption efficiency was determined at various conditions such as adsorbent dosage (10–500 mg/L), sorbate concentrations (100–1500 µg/L), contact time (0–300 min), initial pH (3–10), and the number of recycle.By subtracting the pyrolysis temperature from 450 °C to 900 °C, the surface areas were found to increase from 12.9 m2/g to 353.3 m2/g, respectively.The 143 experimental data were evaluated by a pair of kinetics and isotherm models and the Adaptive Neural Fuzzy Inference System (ANFIS). The developed ANFIS model was 98.56% successful in predicting the CP removal efficiency depending on the adsorption conditions. Walnut Shell Biochar (WSBC) can be applied for CP adsorption with 86.64% removal efficiency under optimum adsorption conditions (adsorbent = 250 µg/L, sorbate = 1000 µg/L, pH = 7.07 and contact time 15 min) thanks to its improved porosity. It was determined that the biochar samples could be reused 5 times. Equilibrium adsorption was observed to conform to the Langmuir isotherm, and the maximum adsorption capacity for WSBC@900 was 3.536 mg/g.  相似文献

Three-dimensional laser Doppler anemometry measurements are performed on developed laminar flow in three helical pipes. The experimental observations are compared to results of numerical calculations employing the fully elliptic numerical method. Good agreement is found between measured data and numerical results. The three helical pipes, with curvature ratios of 0.0734 and 0.1374 and non-dimensional pitches of 0.0793 and 0.193, are adopted to study the effects of curvature and pitch on laminar flow in the experimental approach. The range of Reynolds numbers is 500–2000 to ensure laminar flow in the entire helical pipe. Both the profile shapes of the normal components of the secondary flow and those of the axial flow along the same centerline present not only similar patterns but also similar change when pitch, curvature ratio, and Reynolds number vary. The results demonstrate comprehensive relationships between the axial flow and the secondary flow.  相似文献

A very promising recent trend in applied quantum physics is to combine the advantageous features of different quantum systems into what is called “hybrid quantum technology”. One of the key elements in this new field will have to be a quantum memory enabling to store quanta over extended periods of time. Systems that may fulfill the demands of such applications are comb‐shaped spin ensembles coupled to a cavity. Due to the decoherence induced by the inhomogeneous ensemble broadening, the storage time of these quantum memories is, however, still rather limited. Here we demonstrate how to overcome this problem by burning well‐placed holes into the spectral spin density leading to spectacular performance in the multimode regime. Specifically, we show how an initial excitation of the ensemble leads to the emission of more than a hundred well‐separated photon pulses with a decay rate significantly below the fundamental limit of the recently proposed “cavity protection effect”.

A novel experimental technique is developed for time-resolved detection and tracking of damage in the forms of delamination and matrix cracking in layered materials such as composite laminates. The technique is non-contact in nature and uses dual or quadruple laser interferometers for high temporal resolution. Simultaneous measurements of differential displacement and velocity at individual locations are obtained to analyze the initiation and progression of interfacial fracture and/or matrix cracking/delamination in a polymer matrix composite laminate system reinforced by graphite fibers. The measurements at multiple locations allow the speeds at which interfacial crack front (mode-I) or matrix cracking/delamination front (mode-II dominated) propagates to be determined. Experiments carried out use three-point bend configurations. Impact loading is achieved using a modified Kolsky bar apparatus with a complete set of diagnostics for load, deformation, deformation rate, and input energy measurement. This technique is used to characterize the full process of damage initiation and growth. The experiments also focused on the quantification of the speed at which delamination or damage propagates under primarily mode-I and mode-II conditions. The results show that the speed of delamination (mode-I) or the speed of matrix cracking/delamination (primarily mode-II) increases linearly with impact velocity. Furthermore, speeds of matrix failure/delamination under primarily mode-II conditions are much higher than the speeds of mode-I crack induced delamination under mode-I conditions.  相似文献

本文采用激光颗粒动态分析仪(PDA)测量了钢包底部喷吹气液两相流中气泡的直径和气泡上升速度的分布;采用激光多普勒测速仪(LDA)测量了气液两相区和液体单相循环区液体速度场的分布。测量结果表明:气泡在脱离喷嘴上浮一定距离后,其大小基本保持不变;在气液两相区中,气泡速度和液体速度的分布均服从高斯分布;液体在单相区作循环流动,在侧壁与底部交接处,存在液体流动的“死区”。  相似文献

Recent investigations involving shock waves generation and shock pressure measurement in direct ablation regime and confined ablation regime for aluminium, copper, titanium and steel (40C130) materials are reported. Experimental measurements demonstrated that in direct ablation regime the peak pressures typically are less than 10 Pa when the incident laser intensity is about 10 W/cm and the time duration of the applied pressure is roughly equal to the laser pulse duration. It is shown that confinement of the surface with a transparent overlayer provided an effective method of enhancing laser-induced shock waves pressure in the target material with an order of magnitude for same laser intensity. Also, in this second regime, the pressure is applied over a period much longer than the laser-pulse duration. As an application measurements of the hardness of target surface before and after laser irradiation in direct ablation regime and confined ablation regime are given, and it is shown that the maximum value of surface hardness is obtained in confined ablation regime. We report parametric studies of elastic wave generation by a pulsed laser and associated spalling of thin surface films by the corresponding high stresses. Two different substrate materials, single crystal Si (100) and fused silica, are considered. Spallation behavior of Al thin films is investigated as a function of substrate thickness, film thickness, laser energy, and various parameters governing the source. Surface displacement due to the stress wave is measured by Michaelson interferometry and used to infer the stresses on the film interface. Consistent with previous studies, the maximum stress in the substrate and at the film/substrate interface increases with increasing laser fluence. For many of the conditions tested, the substrate stress is large enough to damage the Si. Moreover, the maximum interface stress is found to increase with increasing film thickness, but decrease with increasing substrate thickness due to geometric attenuation. Of particular significance is the development of a decompression shock in the fused sillica substrates, which results in very high tensile stresses at the interface. This shock enhances the failure of thin film interfaces, especially in thicker samples.  相似文献

The bismuth‐coated electrode is known to be prone to errors caused by copper(II). This study investigates copper(II) interference at bismuth film electrode for the detection of lead(II) and cadmium(II). It was conducted using glassy carbon electrode, while the bismuth film was plated in situ simultaneously with the target metal ions at ? 1200 mV. Copper(II) presented in solution significantly reduced the sensitivity of the electrode, for example there was an approximately 70 % and 90 % decrease in peak signals for lead(II) and cadmium(II), respectively, at a 10‐fold molar excess of copper(II). The decrease in sensitivity was ascribed to the competition between copper and bismuth or the metal ions for surface active sites. Scanning electron microscopy (SEM) and energy dispersive X‐ray (EDX) analysis suggested a large decrease in the amount of bismuth nanoparticles formed on the electrode surface in the presence of copper(II) occurred, validating the competition between copper and bismuth ions for surface active sites. Recovery of the stripping signal of lead(II) and cadmium(II) was obtained by adding ferrocyanide ion to the solution. Finally, the proposed method was successfully applied to determine lead(II) and cadmium(II) in water samples and the method was validated by ICP‐MS technique.  相似文献
